Recently our system administrator have changed the root password of one of the linux servers. the readiness status of the server becomes [Not Ready] with message is [Communication failure between Client xxx.xxx.xxx.xxx and the MediaAgent xxxx.xxx.com. Error returned is:Connection failed. What could be the reason behind this issue? is it network related issue or root password changing issue?

Is Check Readiness working from Client & Commserve?

Check Readiness is not using specific Account/Password.

Can you check that cvping/telnet is working between Client & Media Agent and configure Network Routes as below if needed?
