Do I need to check other Compell Reuse Media options to read them?
Does enabling this option affect the data inside the tape?

Yes - the media cannot be read without this option:
The data on the media is not affected by this operation specifically, but the data can be aged, overwritten etc. Probably a good idea to set infinite retention on the tapes just in case. You can do this after import but before physically having the tapes in the library as a precaution.
